Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: MLK4 (M3KL4) is a protein-serine/threonine kinase of the TKL group and MLK family. It acts as a signaling regulator, implicated in cell survival, inflammation, and apoptosis. MLK4 acts to impair TLR4 signalling, and may activate JNK1/MAPK8, or p38/MAPK14, or ERK2/MAPK1 pathways, although some studies indicate that it may impair MAPK signalling. MLK4 promotes glucose metabolism in specific cancers by increasing glycolysis and regulating PCK1 expression. Homodimerization via the leucine zipper domains is required for autophosphorylation and subsequent activation. It features an N-terminal SH3 domain, a kinase domain, a leucine-zipper region, and a CRIB motif. Mutations in MLK4 have been linked to colorectal cancer (CRC). It is highly expressed in the pancreas and kidney, and its mutation or overexpression is linked to poor prognosis in lung adenocarcinoma and microsatellite stable colorectal cancers.
