Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: MARK4 (Microtubule Affinity-Regulating Kinase 4) is a protein-serine/threonine kinase of the CAMK group and CAMKL family, which regulates microtubule dynamics by phosphorylating microtubule-associated proteins (MAPs), causing them to detach from microtubules. It features an N-terminal catalytic domain, an autoregulatory UBA domain, a spacer region, and a C-terminal tail. It exists in two isoforms: long (MARK4L) and short (MARK4S), with differing expression patterns. It is primarily expressed in the brain and testis. MARK4 is generally found in the cytoplasm, localized at the centrosome and midbody, particularly during cell division. It is associated with the centrosome during mitosis. It is crucial for maintaining microtubule-organizing center (MTOC) function. It phosphorylates tau protein and other MAPs. MARK4 levels are elevated in Alzheimer's disease, contributes to pathological phosphorylation of tau protein. Reduction in MARK4 levels has been shown to protect against diet-induced obesity in mice.
