Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: MARK3 (Microtubule Affinity Regulating Kinase 3; C-TAK1) is a ~110 kDa protein-serine/threonine kinase of the CAMK group and CAMKL family, and regulates cell polarity, microtubule stability, and cell cycle progression. It phosphorylates microtubule-associated proteins (MAP2, MAP4, TAU), reducing their affinity for microtubules, and acts as a key mediator in the Hippo signaling pathway by inhibiting LATS1. It features an N-terminal kinase domain, a UBA (ubiquitin-associated) domain, and a C-terminal spacer. Its activity is controlled by phosphorylation from upstream kinases like LKB1 (serine/threonine-protein kinase STK11) and interaction with 14-3-3 scaffolding proteins. Phosphorylation of Thr-234 and Ser-238 in MARK3 increases phosphotransferase activity. T211A substitution is involved in decreased phosphorylation and activation by LKB1/LKB1 complex. Phosphorylation of Thr-95 and Ser-96 inhibits phosphotransferase activity. MARK3 phosphorylates CDC25C on Ser-216, controlling the G2/M transition of the cell cycle. It also plays a role in regulating the localization of histone deacetylases (HDAC7) and MITF. MARK3 regulates bone mineral density. It is also associated with carcinogenesis, particularly in the pancreas, and has been identified in a form of genetic recessive blindness.
