Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: Frk (Fyn-related kinase) is a protein-tyrosine kinase of the TK group and Src family that is found in the cell nucleus. It is involved in positive regulation of PTEN by increasing its stability through phosphorylation of Tyr-336 and preventing it degradation, and negative regulation of cell proliferation. It may also have functions during G1 and S phase in the nucleus to suppress growth. While it is primarily found in the nucleus, it is also associated with the cytoplasm, extracellular vesicles, and specific granules. FRK is widely expressed in tissues, with high expression levels found in the kidney and duodenum. It may be a tumour suppressor protein (TSP) under certain circumstances. Cancer-related mutations in human tumours point to a loss of function of the protein kinase. Reduced expression has been observed in breast cancer, while mutations are linked to liver cancer (hepatocellular adenomas) and lung cancer. It may act as an oncogene in some cancers, like pancreatic and liver cancers, where its knockdown inhibits tumour growth, highlighting a functional paradox.
