Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: WNK4 (With-No-Lysine Kinase 4; PRKWNK4) is a protein-serine/threonine kinase of the Other group and WNK family. It regulates electrolyte homeostasis. It acts as an integrative regulator of ion transporters, notably the thiazide-sensitive \(\text{NaCl}\) cotransporter (NCC), promoting reabsorption and regulating K+ secretion via ROMK channels. WNK4 can activate the genes SCNN1A, SCNN1B, SCNN1D, SGK1, TRPV5 and TRPV6. The renal potassium channel KCNJ1, through a kinase-mediated mechanism, will be inhibited via clathrin-mediated receptor endocytosis. WNK4 can also phosphorylate NEDD4L. Activation of WNK4 requires autophosphorylation of Ser-335. Phosphorylation of Ser-331 also promotes increased activity. It lacks a conserved lysine in subdomain II of the catalytic domain, and uses a lysine in subdomain I instead. WNK4 is present in the tight junction complex of kidney cells, where it controls paracellular chloride transport and ion permeability. WNK4 activity is regulated by the KLHL3–Cullin-3 E3 ligase complex; impairments in this pathway increase WNK4 abundance, contributing to FHHt.It is highly expressed in the distal convoluted tubule (DCT) and cortical collecting duct (CCD).
