Scientific Background: WNK4 (PRKWNK4) is a protein-serine/threonine kinase of the Other group and WNK family. It regulates electrolyte homeostasis. WNK4 can activate the genes SCNN1A, SCNN1B, SCNN1D, SGK1, TRPV5 and TRPV6. The renal potassium channel KCNJ1, through a kinase-mediated mechanism, will be inhibited via clathrin-mediated receptor endocytosis. WNK4 can also phosphorylate NEDD4L. Activation of WNK4 requires autophosphorylation of S335. Phosphorylation of S331 also promotes increased activity.
