Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: WNK2 (WNK lysine deficient protein kinase 2; PRKWNK2) is a 230-243 kDa protein-serine/threonine kinase of the Other group and WNK family. It plays a critical role in the regulation of electrolyte homeostasis, signalling survival, and proliferation, often via the WNK2-SPAK/OSR1 cascade. Activation requires autophosphorylation of Ser-356. Phosphorylation of Ser-352 also promotes increased phosphotransferase activity. It activates sodium-coupled chloride cotransporters and inhibits potassium-coupled chloride cotransporters. It is also involved in reducing ERK1/2 activation. ERK1/2 expression can also increase through activation of of MEK1 via Rac1-mediate PAK1 activation. Unlike most kinases, WNK2 lacks a catalytic lysine in subdomain II, instead using an alternative lysine in subdomain I for ATP binding. It is predominantly found in the brain and heart, with lower levels in other tissues, including skeletal muscle and kidney. WNK2 may be a tumour suppressor protein (TSP). WNK2 is also called serologically defined colon cancer antigen 43. Loss of WNK2 expression was found in adult gliomas and triggers Rac1-mediated tumour cell invasiveness. Loss of expression of WNK2 is also found in meningioma and facilitates increased cell cycle progression and proliferation.
