Product Type Specific: SIK3 (QSK) protein kinase pan-specific antibody

Antibody Code: NK250-1

Antibody Target Type: Pan-specific

Protein UniProt: Q9Y2K2

Protein SigNET: SIK3

Antibody Type: Polyclonal

Antibody Host Species: Rabbit

Antibody Immunogen Source: Human SIK3 (QSK) sequence peptide Cat. No.: PE-01AYS85

Antibody Immunogen Sequence: MPARIGYYEIDRTIGC

Antibody Immunogen Description: Corresponds to amino acid residues M1 to G15; N-terminus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Antibody Concentration: 0.75 mg/ml

Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol

Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.

Product Use: Western blotting | Antibody microarray

Antibody Dilution Recommended: 2 µg/ml for immunoblotting

Antibody Potency: Medium-strong immunoreactivity with recombinant human SIK3 on protein dot blots.

Antibody Species Reactivity: This antibody detects the target protein in the following species due to conservation of amino acid sequence: Human | Chimpanzee | Mouse | Zebra fish | Fruit fly.

Antibody Positive Control: The observed molecular mass of the processed target protein on SDS-PAGE gels is reported to be around 110-115 kDa.

Antibody Specificity: Very high

Scientific Background: SIK3 (Salt-Inducible Kinase 3; QSK) is a 140 kDa protein-serine/threonine kinase of the CAMK group and CAMKL family. It is a member of the salt-inducible kinase family. It phosphorylates key substrates to manage energy storage, lipid metabolism, and cholesterol synthesis. It also positively regulates mTOR signaling by triggering the degradation of the inhibitor DEPTOR. SIK3 is crucial for homeostatic sleep/wake regulation, where a conserved phosphorylation site (Ser-551 in humans) regulates daily nonrapid-eye movement sleep amounts. It also controls the cell cycle by regulating HDAC4 trafficking. SIK3 is activated by phosphorylation on Thr-163 by STK11 in complex with STE20-related adapter-alpha (STRAD alpha) pseudo kinase and CAB39. It is a AMPK-family kinase that is ubiquitously expressed in the cytoplasm. It is widely expressed in tissues like the liver and brain. SIK3 acts as a tumour-associated antigen and plays complex roles in cancer biology, including facilitating metastatic traits in breast cancer and acting downstream of LKB1 in lung cancer. In ovarian cancer, overexpression of SIK3 promotes G1/S cell cycle progression and cell proliferation. This description may include information annotated by UniProt and/or Google AI.
