Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: SMG1 (Suppressor of Morphogenesis in Genitalia 1; ATX) is a ~410 kDa protein-serine/threonine kinase of the Atypical group and PIKK family. It mediates the genotoxic stress pathway as well as the mRNA surveillance pathway. It is crucial for surveillance and quality control, phosphorylating the UPF1 protein to trigger the degradation of aberrant mRNA transcripts with premature termination codons. SMG1 associates with UPF1 and translation release factors to form the SURF complex, which identifies mRNA mistakes and initiates degradation. SMG1 will target nonsense RNA molecules for degradation via phosphorylation of RENT1/UPF1. SMG1 is essential for optimal phosphorylation and activity of p53/TP53 following genotoxic stress. It features a kinase domain with an extensive N-terminal region and a FATC domain crucial for its kinase activity. SMG1 is essential for embryo development, and its depletion has been linked to increased sensitivity to radiation and cancer development. It is iInhibited by caffeine, LY294002 and wortmannin.
