Scientific Background: SMG1 (ATX) is a protein-serine/threonine kinase of the Atypical group and PIKK family. It mediates the genotoxic stress pathway as well as the mRNA surveillance pathway. SMG1 will target nonsense RNA molecules for degradation via phosphorylation of RENT1/UPF1. SMG1 is essential for optimal phosphorylation and activity of p53/TP53 following genotoxic stress. It is iInhibited by caffeine, LY294002 and wortmannin.
