Product Type Specific: JNK3 (MAPK10 ) protein kinase pan-specific antibody

Antibody Code: NK197-4

Antibody Target Type: Pan-specific

Protein UniProt: P53779

Protein SigNET: JNK3

Antibody Type: Polyclonal

Antibody Host Species: Rabbit

Antibody Immunogen Source: Human JNK3 (MAPK10) sequence peptide Cat. No.: PE-01AXS95

Antibody Immunogen Sequence: MSLHFLYYGEPTLDVC

Antibody Immunogen Description: Corresponds to amino acid residues M1 to D15; N-terminus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Antibody Concentration: 0.25 mg/ml

Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol

Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.

Product Use: Western blotting | Antibody microarray

Antibody Dilution Recommended: 2 µg/ml for immunoblotting

Antibody Species Reactivity: This antibody detects the target protein in the following species due to conservation of amino acid sequence: Human | Dog | Rat | Mouse.

Antibody Positive Control: The observed molecular mass of the processed target protein on SDS-PAGE gels is reported to be around 48-54 kDa.

Antibody Specificity: Very high

Antibody Cross Reactivity: No immunoreactivity on protein dot blots with recombinant human JNK1 and JNK2.

Scientific Background: JNK3 (MAPK10) is a protein-serine/threonine kinase of the CMGC group and MAPK family. It is a neuron-selective kinase primarily involved in stress-induced apoptosis and neurodegenerative diseases. Phosphorylation at Thr-221 and Tyr-223, by MAP2K4 and MAP2K7. MAP2K7, increases phosphotransferase activity. MAP2K7 phosphorylates JNK3 on Thr-221 causing a conformational change and a large increase in Vmax. MAP2K4 then phosphorylates Tyr-223 resulting in a further increase in Vmax. Phosphorylation at Thr-131 inhibits phosphotransferase activity. It is inhibited by dual specificity phosphatases, such as DUSP1 and by HDAC9. Once activated, it phosphorylates transcription factors like c-Jun and ATF2 in the nucleus to regulate gene expression in response to in response to stress signals such as UV radiation, osmotic stress, and inflammatory cytokines. It is highly expressed in the brain, and more weakly in heart, and testes. JNK3 is implicated in promoting neuronal death in neurodegenerative disorders, including Alzheimer’s disease, Parkinson’s disease, and stroke. The near absence of reported somatic mutations in this protein indicates that it may function as a tumour-requiring protein (TRP) in many human tumours.
