Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: JNK2 (MAPK9) is a protein-serine/threonine kinase of the CMGC group and MAPK family. JNK2 is expressed as several isoforms (46 kDa and 54 kDa) through alternative splicing, mainly differing in their C-terminus. It is involved in the control of cell differentiation, transformation, migration, proliferation, and programmed cell death in response to stress and cytokines. JNK2 is known to play a critical role in T-cell differentiation and apoptosis, by preventing ubiquitination of p53 in non-stress cells. Phosphorylation of Thr-183 and Tyr-185 by either of two dual specificity kinases, MAP2K4 and MAP2K7, increases phosphotransferase activity. Phosphorylation of Ser-129 also increases phosphotransferase activity. It is inhibited by dual specificity phosphatases, such as DUSP1, which target the Thr-183 and Tyr-185 activatory phosphorylation sites. JNK2 often functions as a positive regulator of c-Jun, though it can also act as a negative regulator of cell proliferation, unlike JNK1. JNK2 has been linked to several pathologies, including cancer (e.g., lung carcinoma transformation) and inflammation. Studies also indicate its role in promoting cell survival during endoplasmic reticulum (ER) stress. JNK2 has been shown, in a cancer cell line to have a role in breast cancer cell motility.
