Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: PAK6 (PAK5) is a protein-serine/threonine kinase of the STE group and STE20 family. It is primarily expressed in the testis and prostate, in the cytoplasm and nucleus. In some contexts, it acts as a regulator of neuronal, orneurite outgrowth. PAK6 binds to LRRK2 and is involved in neuron projection growth and potential mechanisms related to Parkinson's disease. It plays a role in the regulation of gene transcription. The kinase activity is induced by various effectors including the androgen receptor (AR) or MAP2K6/MAPKK6. PAK6 interacts with AR to regulate prostate cancer growth. It phosphorylates the DNA-binding domain of androgen receptor/AR and inhibits AR-mediated transcription. It also inhibits ESR1-mediated transcription. It may play a role in cytoskeleton regulation by interacting with IQGAP1. It can protect cells from apoptosis through phosphorylation of BAD. The activation of PAK6 through MAP2K6/MAPKK6 can be inhibited through the three mutations S165A, S560A, and Y566F. PAK6 expression is often increased in prostate cancer. PAK6 suppresses tumour progression through regulation of androgen receptor homeostasis.
