Scientific Background: PDK1 (PDPK1) is a protein-serine/threonine kinase of the AGC group and PKB family. It activates, via phosphorylation, many members of the AGC family of protein kinases. It is activated by phosphorylaton at Y9, S160, S241, Y373, and Y376. MELK-mediated phosphorylation can be inhibited through a T354A mutation. PDK1 phosphorylation of MAP3K5 can be ablated through the S394A and S398A mutations. PKB has been observed to be phosphorylated more via a T513E mutation in PDK1. Its phosphotransferase activity can be reduced 3-fold with a A277V mutation. PDK1 is important in mediating migration and metastasis of breast cancer cells.
