Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: MKK6 (MAP2K6, MEK6) is a 37 kDa Dual specificity protein kinase of the STE group and STE7 family. It selectively phosphorylates p38 MAPKs in response to inflammatory cytokines and environmental stresses, regulating crucial processes such as apoptosis, cell cycle arrest, and differentiation. It is critical for mediating inflammatory signalling, cardiomyocyte development, neutrophil migration, and T-cell activation. It can regulate expression of genes critical for survival of newly differentiated neurons. It is activated by dual phosphorylation on Ser-207 and Thr-211; S207E and T211E are linked with constitutive activation. With MAP3K3/MKK3, it catalyzes the concomitant phosphorylation of a threonine and a tyrosine residue in the p38 MAP kinases MAPK11, MAPK12, MAPK13 and MAPK14, and it plays an important role in the regulation of cellular responses to cytokines and all kinds of stresses. Especially, MAP2K3/MKK3 and MAP2K6/MKK6 are both essential for the activation of MAPK11 and MAPK13 induced by environmental stress, whereas MAP2K6/MKK6 is the major MAPK11 activator in response to TNF. MAP2K6/MKK6 also phosphorylates and activates PAK6. Within the p38 MAPK signal transduction pathway, MAP3K6/MKK6 mediates phosphorylation of STAT4 through MAPK14 activation, and is required for STAT4 activation and STAT4-regulated gene expression in response to IL-12 stimulation.
