Production Method: The immunizing peptide was produced on an A.B.I. automated peptide synthesizer. The peptide was cleaved from the synthesis resin with HF and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by amino acid analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: Ksr1 (kinase suppressor of Ras 1) is a 90-100 kDa protein-serine/threonine kinase of the TKL group and RAF family. It is a Raf-related pseudokinase and may directly mediate Raf activation. It acts as a scaffold for the Raf/MEK/ERK kinase cascade. It features five conserved domains (CA1–CA5) and functions mainly as a homodimer, with structural similarities to RAF kinases. It was originally described as a "kinase suppressor" in Drosophila, where its loss suppressed activated Ras mutations. While it possesses a kinase-like domain, its primary function is as a scaffold. Its direct ATP-binding ability is considered low or controversial, but it acts as an allosteric activator of BRAF and facilitates MEK phosphorylation. It resides in the cytoplasm in resting cells and translocates to the plasma membrane upon receptor activation. No specific mutations have been directly implicated in oncogenesis although loss of Ksr1 function will blunt the Ras pathway and therefore attenuate its oncogenic potential.
