Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: TAOk3 (Thousand-and-one amino acid kinase 3; MAP3K18, TAO3) is a protein-serine/threonine kinase of the STE group and STE20 family. It is a cutaneous T-cell lymphoma-associated antigen. It function as a MAP kinase kinase kinase (MAP3K) in mammals, playing crucial roles in immune cell regulation, osteoblast differentiation, dendritic cell terminal differentiation, and cellular stress responses. TAOK3 regulates p38 and JNK cascades, contributes to ADAM10-mediated B-cell development. It also functions in the G2-M transition in response to DNA damage. TAOK3 is found at the cell membrane (membrane rafts), in the cytoplasm, and is associated with lipid droplets, acting as a peripheral membrane protein. Specific single-nucleotide polymorphisms (SNPs) in the TAOK3 gene have been associated with higher opioid requirements for pain management in advanced cancer patients. Insertional mutagenesis studies in mice support a role for this protein kinase in mouse cancer oncogenesis.
