Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ICK (Intestinal cell kinase; CILK1; Ciliogenesis Associated Kinase-1) is a protein-serine/threonine kinase of the CMGC group and RCK family. ICK functions in the regulation of proliferation and differentiation in intestinal cells. It displays sequence similarity to the MAP kinases in the catalytic domain and the requirement for dual phosphorylation for activation within a MAP kinase-like TDY motif. It also features an intrinsically disordered C-terminal domain (CTD), which allows ICK to localize to the base and tip of primary cilia, with its C-terminal domain required for this localization and for interaction with the IFT-B complex. It acts as a key regulator of ciliary length, Hedgehog signaling, and cell cycle progression. It plays a crucial role in ciliogenesis (forming primary cilia) and acts as a kinase for proteins like KIF3A, which regulates cilia transport. ICK homologues in algae and invertebrates function in the formation of flagella and in ciliogenesis. Impairing ICK function has been shown to down-regulate the translation of specific targets of eIF4E and S6K1 (e.g., cyclin D1 and c-Myc) mediated through the mTOR/Raptor signalling pathway. ICK interacts with the mTOR/Raptor complex and phosphorylates Raptor. It is widely expressed across tissues, with high levels in the testis and placenta. Mutations in the gene are associated with endocrine-cerebro-osteodysplasia syndrome (ECO syndrome) and cranioectodermal dysplasia.
