Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: CDK5 is a protein-serine/threonine kinase of the CMGC group and CDK family. It is required for cell cycle arrest in neuronal cells. It regulates neurite outgrowth, neuronal differentiation, and synaptic vesicle recycling. CDK5 is involved in cell motility, survival, apoptosis, and angiogenesis in non-neuronal tissues. It is activated by binding p39 and it truncated p35 protein. Phosphorylation at Ser-159 increases phosphotransferase activity. Phosphorylation at Thr-14 and Tyr-15 inhibits phosphotransferase activity. CDK5 has been observed to be proline-directed and to phosphorylate SRC, NOS3, VIM/vimentin, p35/CDK5R1, MEF2A, SIPA1L1, SH3GLB1, PXN, PAK1, MCAM/MUC18, SEPT5, SYN1, DNM1, AMPH, SYNJ1, CDK16, RAC1, RHOA, CDC42, TONEBP/NFAT5, MAPT/TAU, MAP1B, histone H1, p53/TP53, HDAC1, APEX1, PTK2/FAK1, huntingtin/HTT, ATM, MAP2, NEFH and NEFM.
