Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: CaMK2g is a protein-serine/threonine kinase of the CaMK group and CaMK2 family. It is a calcium/calmodulin-activated kinase that is involved in sarcoplasmic reticulum calcium transport, and may have a role in synaptic plasticity. Once activated by calcium or calmodulin CaMK2g will autophosphorylate at Thr-287, and become activate in the absence of calcium. It mediates autonomous regulation of neuronal dendritic spine formation, sarcoplasmic reticulum transport, and leukemic cell proliferation. It is involved in the JAK-STAT signaling pathway by phosphorylating STAT1, and plays a role in hippocampal-dependent learning and synaptic plasticity. It is highly expressed in the brain, heart, and muscle tissue. It is primarily found in the postsynaptic density (PSD) of neurons and the cytoplasm, but also within the plasma membrane and nucleus. CaMK2g resides in large multi-oligomeric protein complexes (12 subunits) with other CaMK2 subunits (alpha, beta, and delta). It is implicated in autosomal dominant intellectual developmental disorder 59 (MRD59). It is studied as a factor in cancer cell proliferation and vascular smooth muscle migration.
