Antibody Immunogen Description: Corresponds to amino acid residues C7 to S26; N-terminus and kinase domain I, highly conserved in all CaMK2 isoforms.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: CaMK2d is a protein-serine/threonine kinase of the CaMK group and CaMK2 family. It is a calcium/calmodulin-activated protein kinase. CaMK2d resides in large multi-oligomeric protein complexes (12 subunits) with other CaMK2 subunits (alpha, beta, and gamma). It is activated in response to a rise in intracellular Ca(2+) concentration. Autophosphorylation at Thr-287 probably increases phosphotransferase activity in a Ca(2+)-independent manner. It is primarily expressed in the heart and brain, that regulates calcium signaling, cardiac muscle contraction, and excitation-contraction coupling. It mediates cardiac stress responses (hypertrophy, heart failure) and neuronal function, with overactivation linked to diseases. It phosphorylates proteins involved in cardiac function, such as ryanodine receptors, phospholamban, and L-type channels, enhancing cardiac performance under stress. Overexpression or aberrant activation of CaMK2d is a major driver of cardiac pathology, including myocardial infarction, cardiac hypertrophy, and arrhythmias (dilated cardiomyopathy).
