Scientific Background: Kv7.2 (KvLQT2) (encoded by the KCNQ2 gene) is a voltage-gated potassium channel protein crucial for regulating neuronal excitability. It forms homo- or heterotetramers (mainly with Kv7.3) to produce the "M-current," a low-threshold, non-inactivating current that prevents neuronal hyperexcitability. Kv7.2 features six transmembrane segments (S1-S6), with a voltage-sensing domain (S1-S4) and a pore domain (S5 & S6). A large C-terminus is vital for assembly and trafficking. It becomes activated at subthreshold potentials (≥ -60 mV)) to stabilize the resting membrane potential. It requires phosphatidylinositol-4,5-bisphosphate (PIP2) to open the channel. Kv7.2 is highly expressed in the developing and adult brain, primarily found in neuronal cell bodies. KCNQ2 mutations are linked to Benign Familial Neonatal Convulsions (BFNC) (1) and early-onset epileptic encephalopathy.
