Scientific Background: CACNB1 (Calcium Voltage-Gated Channel Auxiliary Subunit Beta 1) is a critical regulatory protein, specifically the b1 subunit of the L-type calcium channel (dihydropyridine receptor) in skeletal muscle. It binds to the pore-forming a1S subunit to promote membrane targeting, increase calcium current, and modulate voltage activation, playing a key role in excitation-contraction coupling. It plays an important role in the calcium channel by modulating G protein inhibition, increasing peak calcium current, controlling the alpha-1 subunit membrane targeting and shifting the voltage dependence of activation and inactivation. CACNB1 is heavily expressed in skeletal muscle and cardiac tissues, and it is primarily localized in the T-tubules and sarcolemma of skeletal muscle cells. It increases the peak calcium current and manages the trafficking of the channel complex to the plasma membrane.
