Scientific Background: Aquaporins selectively conduct water molecules in and out of the cell, while preventing the passage of ions and other solutes. Known as water channels, they are integral membrane pore proteins (1, 2). Aquaporin-4 (AQP4) is the primary water-transporting protein in the mammalian central nervous system (CNS), forming highly selective bidirectional channels in the plasma membrane of astrocytes, particularly at the blood-brain barrier. AQP4 is up regulated by direct insult to the CNS (4). It facilitates rapid water movement, maintaining brain water homeostasis, AQP4 is constitutively expressed in the basolateral cell membrane of principal collecting duct cells in the kidney and provide a pathway for water to exit these cells (3). AQP4 is a homotetramer, consisting of four identical subunits, with a molecular mass of roughly 34-38 kDa per monomer. It exists as a transmembrane protein with six helical, membrane-spanning domains and two short helical segments creating a narrow, selective aqueous pore of ~2.8 Å in diameter.
