Antibody Code: NK104-4

Antibody Target Type: Pan-specific

Protein UniProt: Q13163

Protein SigNET: MEK5

Antibody Type: Polyclonal

Antibody Host Species: Rabbit

Antibody Immunogen Source: Human MEK5 (MAP2K5, MKK5) sequence peptide Cat. No.: PE-01AVZ99

Antibody Immunogen Sequence: CEELMGHPFIVQFNDG

Antibody Immunogen Description: Corresponds to amino acid residues E411 to G425; Kinase last alpha-chain.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Antibody Concentration: 1 mg/ml

Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol

Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.

Product Use: Western blotting | Antibody microarray

Antibody Dilution Recommended: 1 µg/ml for immunoblotting

Antibody Species Reactivity: This antibody detects the target protein in the following species due to conservation of amino acid sequence: Human | Chimpanzee | Rhesus Macaque | Rat | Mouse.

Antibody Positive Control: The observed molecular mass of the processed target protein on SDS-PAGE gels is reported to be around 47-52 kDa.

Antibody Specificity: High

Antibody Cross Reactivity: No immunoreactivity on protein dot blots with recombinant human MEK1, MKK4 and MKK6.

Scientific Background: MEK5 (MAP2K5) is a Dual specificity protein kinase of the STE group and STE7 family. It is essential for early embryogenesis, cell survival, angiogenesis, cardiovascular development, and vascular function. It is activated by phosphorylation at Ser-311 and Thr-315 by MAP kinase kinase kinases such as MAP3K2/3 (MEKK2/3) or atypical protein kinase C (aPKC) isoforms. Activation of this pathway appear to play a critical role in protecting cells from stress-induced apopotosis, neuronal survival and cardiac development and angiogenesis by acting as a scaffold protein for the construction of a complex consisting of MAP3K2 or MAP3K3 and MAP3K5 and MAPK7. Alternative splicing produces two isoforms, with MEK5-alpha containing a N-terminal PB1 (phox and Bem1p) domain that binds to ERK5 and acts as a scaffold for the signalling complex, which is essential for mediating interactions with upstream activators. Overexpression and overactivation of the MEK5/ERK5 pathway are associated with various cancers (e.g., prostate cancer), promoting tumour growth and chemotherapy resistance.
